The Drosophila P-element transformation usually involves the injection of transposon-based construct with the white and/or the yellow marker to w1118 or yw host line. We perform injection with your (or our) purified plasmid construct (helper P-element plasmid will be supplied by us unless you prefer to use your own) and return you the injected lavae, or the stable lines and/or the balanced lines depending on your chosen service.
PhiC31 Integrase-mediated Transgenesis Systems
PhiC31 integrase-mediated transgenesis systems are based on the site-specific bacteriophage PhiC31 integrase which mediates sequence-directed, irreversible and highly efficient integration between a bacterial attachment site (attB) and a phage attachment site (attP). We have the P{CaryP}, PhiC31-RMCE, P[acman], FlyC31, P{CaryIP} system’s host lines listed in BDSC, and we can prepare MiMIC lines from BDSC for you. We will inject the attB construct into the chosen attP docking site embryos and return you the injected larvae, the stable lines without the PhiC31 source and/or the balanced lines depending on your chosen service.
Genome Engineering (CRISPR) Transformation
The Drosophila CRISPR/Cas9 system is a powerful genome engineering tool that are composed of programmable, sequence-specific DNA-binding modules associated with a nonspecific DNA nuclease, which induce DSB (DNA double-strand break) that stimulate NHEJ (non-homologous end joining) or HR (homologous recombination) repairing mechanisms at specific genomic location to introduce indel, mutation or fragment replacement.
We provide other injection except the above services according to your experimental needs. Usually piggyBac, Minos and other injectable materials are categorized here. We will inject your sample into your selected host line and return you the injected larvae, the stable lines and/or the balanced lines depending on your chosen service.
We also provide related molecular lab services, e.g. DNA preparation (Qiagen Midiprep), BAC preparation (Qiagen Large Construct Maxiprep), transposable element mapping for P-element lines, molecular mapping for CRISPR HDR lines, PCR confirmation of recombination site, orientation screening for MiMIC lines, etc.
